Histochemical Studies on the Activity of ATPase and Autoradiographic Study on the Uptake of 14C ?ATP in Ascaris Lumbricoides Suis


Abstract
I) Histochemical studies were carried out in order to identify the distribution of ATPase in A>scaris lumbricoides suis.
The worms s were cultivated in the incubator, keeping at 37 0C Ringer-Dale solu¡þtion, for 3 days.Thereafter they were immersed in cold acetone solution and kept in a freezer at-70oC.The worms were sectioned by cryostat preparation to 8.0¥ì in thickness and stained by Padvkula-Herman method. The result was obtained as follow; microscopically the ATPase was found in the subcuticuarsyncytial cells, strial zone of the epithelium of the intestine, and reticular cells.
2. A microautoradiographic study was performed in order to know the uptake of 14C-ATP
by Ascaris lumhrlcoldes suis.
The worms were kept in tyrode solution containing 14C?ATP for one hour at 370C. After the incubation , the worms were sectioned in cryostat and applied the technique of Fitzgerald stripping microautoradiography.
As the results, the black fine granules which were derived from the labeled 14C?ATP substances were highly concentrated in the epithelial cells of the intestine and the subcuticular tissues of Ascaris Lumbricoids suis.
